Nijmegen Proteomics Facility

Institution / Head office:
University Medical Centre Nijmegen
Summary:

The Nijmegen Proteomics Facility (NPF) joins all proteomics activities of the Radboud University Nijmegen and the Radboud University Medical Centre. The NPF has equipment and experience for proteomics activities at its disposal.

Description:

The Nijmegen Proteomics Facility has central proteomics facilities at its disposal which are accessible for all groups within the Radboud University Nijmegen, Radboud University Nijmegen Medical Centre and outside the centre. The Nijmegen Proteomics Facility is a virtual laboratory spread all over the campus of the Radboud University Nijmegen.

Facilities available are:

Fujifilm FLA-5100 fluorescent image analyzer

Category: Fluorescence and Radioisotopic Imaging

Description:
The FLA-5100 imaging system allows imaging of fluorescent and radioisotopic samples as large as 40 x 46cm at pixel size as low as 10 microns. This system is especially suitable for protein electrophoresis in the proteomics field and general array analysis for genomics. The FLA-5100 is ideal for fluorescence detection of 2D gel by SYPRO® Ruby stain, fluorescence detection of CBB by 670nm laser, and digitization of CBB-stained or silver-stained samples.

Specifications:
Lasers: 473 nm, 532nm, 635 nm
Dynamic range: five orders of magnitude
Pixel size: user choice or 10-25-50-100-200 micron
Filters: /Y510, /O575, /R665, 530DF20 + /665, 570DF20 + /665, 508FS10-25

nLC LIT FT-ICR Mass Spectrometer

Category: Proteomics

Description:
Nanoflow Liquid Chromatography (nLC) is used for its high resolution separations and high sensitivity. In this system nLC is combined with a Linear Ion Trap Fourier-Transform Ion Cyclotron Resonance mass spectrometer (LIT FT-ICR MS). This hybrid instrument combines LIT and FT-ICR technologies into a single instrument with unprecedented analytical power and versatility. Ultra high resolution and sensitivity coupled with ppm mass accuracy, and MSn are universally available for the toughest applications demanding rigorous structural characterization. Protein identification in highly complex mixtures can be performed.

Specifications:
Mass range: 50-4000 m/z
Resolution: 100,000 at m/z 400 at 1 s per scan (max res. >500,000 at slower rates)
Mass accuracy: < 2 ppm with external calibration
Sub fmol sensitivity
Available fragmentation modes: CID, IRMPD, ECD
Electrospray ionisation

Lab with CCKL-accreditation

Availability:
This instrument requires well trained specialists to operate and it is therefore only available for measurements performed by the associated specialist. Different proteomics services are offered (analysis is performed by operators of the facility). For large projects (min. 1 day/week for 1 year), the instrument is available to the customer after an extensive training by the associated specialist. For more information on services and possibilities, please contact Jolein Gloerich.

vMALDI-LIT MS

Category: Proteomics

Description:
The vMALDI ion source system provides fast, automated MS and Data Dependent MS/MS analysis of digested protein mixtures in a single, rapid experiment. While LC/MS analysis times are typically 30 to 60 minutes, using the Finnigan LTQ with the vMALDI ion source generally requires less than a minute per sample enabling high throughput protein identification. This integrated system combines the speed of MALDI with the sensitivity of the LTQ to create Mascot-searchable MS/MS spectra for confident protein assignments. This system is ideally suited for high-througput protein identification from 2D-gel electrophoresis experiments.

Specifications:
Mass range: 50-4000 m/z
MSn (n=1-9)
Resolution:
Mass accuracy: 500 ppm
fmol sensitivity
Available fragmentation modes: CID, PQD
MALDI ionisation

Lab with CCKL-accreditation

Availability:
This instrument requires well trained specialists to operate. For small projects, different proteomics services are offered (analysis is performed by operators of the facility). For large projects (min. 1 day/week for 0.5 year), the instrument is available to the customer after an extensive training by the associated specialist. For more information on services and possibilities, please contact Jolein Gloerich.
